Abstract: Teratogenicity was evaluated in pregnant female Sprague Dawley albino rats (24/group) orally exposed by gavage to ((3,5-bis(1,1- dimethylethyl)-4-hydroxyphenyl)methylthio)acetic acid, 2-ethylhexyl ester (IRGANOX 1192) at dose levels of 0, 50, 150 or 300 mg/kg/day on gestation days (GD) 6-15. Surviving animals were sacrificed on GD 21. Significant differences were observed between treated and control animals in the following: decreased food consumption and body weight gain (high-dose group), increased resorption rate (high-dose, embryonic deaths), increased incidence of ectrodactyly (high-dose, variable number of limbs concerned), and increased incidence of delay of ossification (high-dose group, mainly in phalangeal nuclei and calcanei in the distal limbs). No significant differences were observed between treated and control animals in the following: maternal clinical signs of toxicity, or mortality, external or macroscopic examination of maternal organs, pregnancy rate, corrected body weight gain, numbers of implantation sites, abortions, and live or dead fetuses, fetal sex ratio or body weight, and visceral examination.
